Product Overview

Composition

The active substance is baclofen 10 mg;

Excipients - lactose, potato starch, gelatin, talc, magnesium stearate, ethyl cellulose.

pharmachologic effect

Baklosan - antispastic, muscle relaxant.

Pharmacodynamics

Reducing the excitability of the terminal sections of afferent sensory fibers and suppressing intermediate neurons, inhibits mono- and polysynaptic transmission of nerve impulses; reduces prestressing of muscle spindles. It has no effect on neuromuscular transmission. With neurological diseases, accompanied by spasticity of skeletal muscles, weakens painful cramps and clonic convulsions; increases the range of motion in the joints, facilitates passive and active kinesitherapy (physical exercises, massage, manual therapy).

Pharmacokinetics

After ingestion, Cmax in plasma is reached after 2-3 hours. Absorption is high. Protein binding is 30%. Penetrates through the placental barrier, excreted in breast milk. Metabolized in the liver. It is excreted by the kidneys (mainly unchanged).

Indications

  • Increased muscle tone in multiple sclerosis; 
  • diseases of the spinal cord of infectious, degenerative and traumatic origin (for example, syringomyelia tumors, motor neuron diseases, trauma); 
  • a stroke; 
  • cerebral palsy; 
  • meningitis; 
  • head injuries; 
  • alcoholism (affective disorders).

Contraindications

  • psychoses
  • epilepsy
  • Parkinson's disease
  • hypersensitivity to the drug Baklosan.

Side effects

Nausea, vomiting, drowsiness, dizziness, increased fatigue, dry mouth, confusion, gait disturbance, urinary retention, constipation or diarrhea, decreased blood pressure, apathy; euphoria, depression, paresthesia, myalgia, muscle weakness, ataxia, tremor, nystagmus, paresis of accommodation, hallucinations, dysuria, enuresis, convulsions, decreased seizure threshold.

With prolonged use - impaired renal function.

Interaction

Enhances the effect of drugs that affect the central nervous system, antihypertensive drugs, ethanol. Tricyclic antidepressants enhance a decrease in muscle tone; levodopa and cardidopa increase the risk of developing hallucinations, confusion, agitation.

Patients should inform their doctor about all medications they are taking.

How to take, course of administration and dosage

Inside, while eating. If you miss the next dose, you should not take a double dose of the drug.

Adults: initial dose of 5 mg (1/2 tablet of 10 mg) 3 times a day. The dose of the drug is gradually increased every 3 days until the therapeutic effect occurs (usually up to 30 - 75 mg per day). In patients requiring higher doses of Baclofen (from 75 mg to 100 mg per day), the drug should be used in 25 mg tablets. In patients over 65 years of age, the dose should be increased with caution in connection with an increased risk of side effects. The maximum daily dose is 100 mg.

Children: the initial dose of 5 mg (1/2 tablet of 10 mg) 3 times a day. If necessary, the dose can be increased with caution every 3 days. The following doses are usually recommended: from 1 to 2 years - 10 - 20 mg per day, from 2 years to 6 years - 20 -30 mg per day, from 6 to 10 years - 30 - 60 mg per day. In children over 10 years, the maximum dose is 1.5 - 2.0 mg / kg body weight.

Overdose

Symptoms: muscle hypotension, depression of the respiratory center, confusion, coma (after returning consciousness, muscle hypotension can persist for 72 hours).

Treatment: heavy drinking, diuretics, with respiratory depression - mechanical ventilation. There is no specific antidote.

Special instructions

In patients with liver diseases and diabetes mellitus, it is necessary to periodically monitor the activity of “liver” transaminases, alkaline phosphatase, and blood glucose. During treatment, one should refrain from potentially hazardous activities associated with the need for increased attention and quick mental and motor reactions.
